主要研究领域及兴趣:
真核细胞发育和分化过程中,功能基因组的表达受到精细而广泛的调控,主要通过相应的调控元件如启动子增强子等实现。近年来的研究发现,真核细胞内染色体组蛋白的特定修饰与基因调控元件密切相关,如H3K4me3特异性标记启动子,H3K4me特异性标记增强子,而H3K27ac标记活化的调控原件。通过组蛋白特定修饰相对应的抗体进行高通量的Chip-seq实验,在全基因组范围内揭示功能基因组的调控元件,并结合生物信息学的方法统计分析与调控元件相结合的转录因子,从而确定真核细胞发育和分化的关键基因及其调控网络。系统性的研究生命过程中功能基因组表达和相互之间的调控关系,从而绘制基因调控回路,全面解析生命过程。
In the eukaryotic cell development and differentiation, functional genes are widely regulated in an exactly pattern, through various regulatory elements such as promoters and enhancers. Considering chromosome chemical structures, histone modifications are highly correlated with the regulatory elements. H3K4me3, H3K4me and H3K27ac specifically label promoters, enhancers and active elements, respectively. Genome wide regulatory elements are revealed via high through-put Chip-seq experiments, and binding transcription factors are revealed through bio-informatics. We will figure out the key genes and regulatory networks in eukaryotic cell development and differentiation, study functional gene patterns and dissect life processes systematically.
